1-(2,5-Dioxo-2,5-dihydro-1H-pyrrol-1-yl)-2-oxo-6,9,12,15,18,21,24,27-octaoxa-3-azatriacontan-30-oic acid Tert-Butyl 1-amino-3,6,9,12,15,18,21,24-octaoxaheptacosan-27-oate (100 mg, 201 mumol) was initially charged in 1.0 ml of DMF and (2,5-dioxo-2,5-dihydro-1H-pyrrol-1-yl)acetic acid (46.8 mg, 301 mumol), 1-hydroxy-1H-benzotriazole hydrate (76.9 mg, 502 mumol) and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (77.0 mg, 402 mumol) were added. The reaction mixture was stirred overnight at RT and then ethyl acetate was added. The organic phase was washed twice with 5% citric acid solution, with sat. sodium hydrogen carbonate solution and then with sat. sodium chloride solution. The organic phase was dried over magnesium sulfate. The solvents were evaporated under vacuum and the residue purified by prep. RP-HPLC (column: Reprosil 125*30; 10mu, flow: 50 mL/min, MeCN/water/0.1% TFA). The solvents were evaporated under vacuum and the residue dried under high vacuum. This gave 19.1 mg (13% of theory) of the compound tert-Butyl 1-(2,5-dioxo-2,5-dihydro-1H-pyrrol-1-yl)-2-oxo-6,9,12,15,18,21,24,27-octaoxa-3-azatriacontan-30-oate.
