ALKALOIDS FROM THE ROOTS OF Papaver pseudo-orientale (FEDDE) MEDW.

Two new alkaloids, pseudorine (Ia) (quaternary benzyltetrahydroisoquinoline type) and pseudoronine (II) (benzil type), were isolated from the roots of Papaver pseudo-orientale (FEDDE) MEDW.This is a first occurence of this type of alkaloids in Papaveraceae.The main component of the tertiary alkaloid fraction was isothebaine accompanied by smaller amounts of orientalidine, mecambridine, bracteoline, nuciferine, protopine, and allocryptopine.The presence of salutaridine and papaverrubine D was established.Traces of coptisine and palmatine were also found in some samples.Strongly polar portion of the extract was converted to iodides.N-Methylisothebainium iodide (IIIa) (major component), pseudorine iodide (Ia), and corytuberine hydriodide (IV) (significant amounts), a minor alkaloids such as alborine iodide (alkaloid PO-5), pseudoronine (II), and magnoflorine iodide (IIIb) were isolated from this source.Structure of pseudorine was determined by UV, IR, 1H, and 13C NMR spectroscopy and using the correlation with laudanosine and codamine.Proposed structure of pseudoronine is based on the mass spectra comparison.

A 70% ethanol extract of California poppy (Eschscholzia californica) was able to bind to 5-HT1A and 5-HT7 receptors at 100 mug/mL. The subsequent isolation procedure yielded the known alkaloids californidine (1), escholtzine (2), N-methyllaurotetanine (3), caryachine (4), and O-methylcaryachine (5), along with a new pavine alkaloid, 6S,12S-neocaryachine-7-O-methyl ether N-metho salt (7). The structure of 7 was determined by spectroscopic data interpretation, while the absolute stereochemistry was determined by means of circular dichroism. From the results obtained from the radioligand-binding assay of the pure compounds, including the commercially available protopine (6), it was evident that the activity on the 5-HT1A receptor was at least partly due to the presence of the aporphine alkaloid 3, which showed the highest inhibition of [3H]8-hydroxy-2-(di-N-propylamino)tetralin ([3H]8-OH-DPAT) binding with an EC50 value of 155 nM and a Ki of 85 nM.

Alzheimer’s disease is an age-related, neurodegenerative disorder, characterized by cognitive impairment and restrictions in activities of daily living. This disease is the most common form of dementia with complex multifactorial pathological mechanisms. Many therapeutic approaches have been proposed. Among them, inhibition of acetylcholinesterase, butyrylcholinesterase, and prolyl oligopeptidase can be beneficial targets in the treatment of Alzheimer’s disease. Roots, along with aerial parts of Argemone platyceras, were extracted with ethanol and fractionated on an alumina column using light petrol, chloroform and ethanol. Subsequently, repeated preparative thin-layer chromatography led to the isolation of (+)-laudanosine, protopine, (-)-argemonine, allocryptopine, (-)-platycerine, (-)-munitagine, and (-)-norargemonine belonging to pavine, protopine and benzyltetrahydroisoquinoline structural types. Chemical structures of the isolated alkaloids were elucidated by optical rotation, spectroscopic and spectrometric analysis (NMR, MS), and comparison with literature data. (+)-Laudanosine was isolated from A. platyceras for the first time. Isolated compounds were tested for human blood acetylcholinesterase, human plasma butyrylcholinesterase and recombinant prolyl oligopeptidase inhibitory activity. The alkaloids inhibited the enzymes in a dose-dependent manner. The most active compound (-)-munitagine, a pavine alkaloid, inhibited both acetylcholinesterase and prolyl oligopeptidase with IC50 values of 62.3 ¡À 5.8 muM and 277.0 ¡À 31.3 muM, respectively.

Acetylcholinesterase inhibitors from the aerial parts of Corydalis speciosa.

In a bioassay-guided search for acetylcholinesterase inhibitors from Korean natural resources, four isoquinoline alkaloids, corynoxidine (1), protopine (2), palmatine (3), and berberine (4) have been isolated from the methanolic extract of the aerial parts of Corydalis speciosa. Structures of these compounds were elucidated on the basis of spectroscopic techniques. These compounds inhibited acetylcholinesterase activity in a dose-dependent manner, and the IC50 values of compounds 1-4 were 89.0, 16.1, 5.8, and 3.3 microM, respectively.

Clarifying the correlation between the chemical structure of mechanophores and their mechanical reactivity informs the design of mechanochemical systems. One specific correlation that has received much recent attention is that between stereoisomerism and mechanical reactivity. Here, we report previously unobserved differences in the mechanical reactivity of furan-maleimide Diels-Alder (DA) stereoisomers. We evaluated the internal competition between the mechanically triggered retro-DA reaction and the mechanochemical ring opening of gem-dichlorocyclopropane mechanophores in the pulsed sonication of polymer solutions. The relative extent of the two sonomechanochemical reactions in the same polymer shows that the endo DA isomer exhibits greater mechanical lability than its exo isomer. This result contrasts with recent measurements of the relative rates of scission in a similar system and points to potential enhanced sensitivity obtained through the use of internal competition as opposed to absolute rates in assessing mechanical reactivity in sonication studies.

Here, we report the discovery of a series of covalent GSK-3beta inhibitors by optimizing both non-covalent interactions and reactive groups. Among these covalent inhibitors, compound 38b with a mild alpha-fluoromethyl amide reactive group emerges as a selective and covalent inhibitor against GSK-3beta, effectively inhibits the phosphorylation of glycogen synthase and tau protein, and increases beta-catenin’s levels in living cells. In addition, compound 38b is highly permeable and not a substrate of P-glycoprotein.
