Effect of Particle Wettability and Particle Concentration on the Enzymatic Dehydration of n-Octanaloxime in Pickering Emulsions

Pickering emulsion systems have emerged as platforms for the synthesis of organic molecules in biphasic biocatalysis. Herein, the catalytic performance was evaluated for biotransformation using whole cells exemplified for the dehydration of n-octanaloxime to n-octanenitrile catalysed by an aldoxime dehydratase (OxdB) overexpressed in E. coli. This study was carried out in Pickering emulsions stabilised solely with silica particles of different hydrophobicity. We correlate, for the first time, the properties of the emulsions with the conversion of the reaction, thus gaining an insight into the impact of the particle wettability and particle concentration. When comparing two emulsions of different type with similar stability and droplet diameter, the oil-in-water (o/w) system displayed a higher conversion than the water-in-oil (w/o) system, despite the conversion in both cases being higher than that in a classic two-phase system. Furthermore, an increase in particle concentration prior to emulsification resulted in an increase of the interfacial area and hence a higher conversion.

Road performance of liquid nitrile-butadiene rubber modified unsaturated polyester resin concrete

Unsaturated polyester resin concrete (UPC) is currently gaining attention in many fields due to its functional compressive capacity and corrosion resistance. However, the toughness of UPC is poor due to the weak resistance to the deformation of the cured unsaturated polyester resin (UP). In this paper, the UP was modified with liquid nitrile-butadiene rubber (LNBR) and further used to make concrete. At the same time, two different initiators were used to cure the UPC and LNBR modified UPC. To study the mechanism and road performance of LNBR modified UPC performance, optical microscopy test, Marshall test, water immersion Marshall test, rutting test, freeze-thaw split test, and infrared spectroscopy experiments were carried out. Test results showed that LNBR could effectively improve the low-temperature toughness of UPC. Although LNBR slightly reduced the Marshall stability and rutting resistance of UPC, it hardly changed the water stability of the UPC. The whole-cell nitrilase-catalyzed asymmetric hydrolysis of nitriles is a green and efficient preparation approach for chiral carboxylic acids, but often suffers from toxicity and cell lysis from organic substrates. In this work, a novel integrated process for whole-cell nitrilase-catalyzed asymmetric hydrolysis was developed for the first time by introducing a biocompatible ionic liquid (IL)-based biphasic system. The whole-cell nitrilases displayed an outstanding stability and recyclability in the biphasic system and still retained > 85% activity even after 7 cycles reaction. A preparative-scale fed-batch hydrolysis of o-chloromandelonitrile to (R)-o-chloromandelic acid (RCMA) was performed using the integrated process. The results revealed a yield of 91.3% and a space-time yield of 746.4 g.L-1.d(-1), which are currently the highest reported values for R-CMA biosynthesis. The proposed integrated process avoids substrate inhibition, facilitates the reusability of whole-cell nitrilases, and thus shows great potential for the sustainable production of chiral carboxylic acids.
