Multicolor Activatable Raman Probes for Simultaneous Detection of Plural Enzyme Activities

Raman probes based on alkyne or nitrite tags hold promise for highly multiplexed imaging. However, sensing of enzyme activities with Raman probes is difficult because few mechanisms are available to modulate the vibrational response. Here we present a general strategy to prepare activatable Raman probes that show enhanced Raman signals due to electronic preresonance (EPR) upon reaction with enzymes under physiological conditions. We identified a xanthene derivative bearing a nitrile group at position 9 (9CN-JCP) as a suitable scaffold dye, and synthesized four types of activatable Raman probes, which are targeted to different enzymes (three aminopeptidases and a glycosidase) and tuned to different vibrational frequencies by isotope editing of the nitrile group. We validated the activation of the Raman signals of these probes by the target enzymes and succeeded in simultaneous imaging of the four enzyme activities in live cells. Different cell lines showed different patterns of these enzyme activities.

Electron Paramagnetic Resonance (EPR) was used to investigate the Tempyo spin label (3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-yloxy) as a report group for the interactions and the conformational changes of lyophilized bovine serum albumin (BSA) and bovine hemoglobin (BH), as function of pH values in the range 2.5-11. The EPR spectra are similar with those of other non-covalently spin label porphyrins in frozen solution at very low temperatures. This behavior indicated a possible spin-spin interaction between the hemic iron and the nitroxide group. The changes in the EPR spectra as function of the pH are discussed in terms of conformational changes of the proteins. Spectral simulations and magnetic EPR parameters reveal the following: (i) one single paramagnetic species, with Gaussian line shape, was used for the best fits of experimental spectra in the case of serum albumin samples; and (ii) a weighted sum of Lorentzian and Gaussian line shape in the case of hemoglobin samples. The representation of correlation time vs. pH, reveals a dependence of degree of immobilization of spin label on the conformational changes of proteins in acidic and basic environment. With the deepening of the concept of green tires, the effective composition of tire rubberis directly related to the qualification of rubber. But tire rubber is a black analysis system for the inspection department, and it’ sexceedingly crucialto accurately detect rubber components by the existing methods. Terahertz time-domain spectroscopy (THz-TDS) technology has been successfully applied to material detection and analysis, but the terahertz spectral data observed from a complex sample of rubber represents the comprehensive results of several interrelated components or interaction of characteristic components in many cases, where as the actual information contained in the raw data may overlap, which will conversely affect the analysis of the components in the rubber mixture. In order to solve the problem of terahertz spectral overlap, the characteristics of continuous smoothing of terahertz spectral matrix and sparse concentration matrix are combined this paper, then the 2 norms with smoothing characteristics and the 1/2 norm with sparsity characteristics into the non-negative matrix factorization method is introduced, which are applied to the separation of terahertz aliased spectra, so as eparation method of terahertz aliasing spectral based on spectral feature Constrained Non-negative Matrix Factorization (CNMF) is proposed. Firstly, nitrile-butadiene rubber combined with vulcanization accelerator 2-Mercaptobenzothizzole(MBT) to form a binary mixture in diverse proportions, and it combined with vulcanization accelerators MBT and tetramethyl thiuram monosulfide (TMTM) to form a ternary mixture in different proportions. Then the terahertz time domain spectrum of all samples ismeasured by terahertz spectroscopy system, which the measured data is subjected too btain a corresponding absorbance spectrum. Further, principal component analysis is performed on the obtained spectral matrix to initially determine the number of components of the mixture. Finally, the Non-negative Matrix Factorization (NMF), Non-negative Matrix Factorization based on pure variables initialization(PNMF) and CNMF methods are used to the decomposition of the mixture data matrix and spectral analysis of the aliased spectrum. The results show that the separation effect of the CNMF algorithm is better than that of NMF and PNMF method, and the corresponding results of the characteristic absorption peak are accurate. In addition, the correlation coefficients of separation results for different component mixtures are higher than 89%, and the spectral angles are less than 0.5 with a higher reduction degree of purity spectrum. Therefore, the constrained non-negative matrix factorization algorithm is introduced into the separation of terahertz aliasing spectra, which is preferable to extract the characteristic information of single components in complex mixtures and provides a better foundation for the qualitative analysis and quantitative calculation of subsequent terahertz multicomponent mixtures as well as the considerable research prospects in the field of quality testing of green tires and rubber.

The exploration of highly-efficient oxygen evolution reaction (OER) electrocatalyst with well-defined structure to understand specific structure-performance relationship is crucial to meet the requirement of water-splitting. Herein, we firstly used atomically dispersed IrClx to uniformly decorate ultrathin Co(OH)(2) nanosheets to dramatically improve electrocatalytic activity. The synthesized IrClx-Co(OH)(2) nanosheets/carbon nanotubes (CNTs) exhibits an overpotential of 230 mV to reach 10 mA/cm(2), which was much enhanced compared to that of pristine Co(OH)(2)/CNTs (308 mV) and commercial benchmark IrO2(309 mV). X-ray absorption fine structure and density functional theory simulations demonstrate strong interfacial interaction between IrClx and Co(OH)(2) nanosheets via the Cl-Ir-O and Ir-Cl-Co bond can efficiently boost its electronic conductivity. The accelerated charge transfer promotes the formation of more positively charged O atoms around cobalt centers, which is beneficial for the deprotonation on IrClx-Co(OH)(2) and makes the catalyst facilitate OER.
