This study describes a benign C-H cyanation of terminal alkynes with alpha-cyanoesters serving as a nontoxic cyanide source. In situ generation of the key copper cyanide intermediate is proposed by a sequence of alpha-C-H oxidation and copper-mediated beta-carbon elimination of alpha-cyanoesters, releasing the alpha-ketoester byproduct observed experimentally. The ensuing reaction of copper cyanide with terminal alkynes delivers preferentially cyanoalkynes and surpasses the possible Glaser type dimerization of terminal alkynes or the undesired accumulation of HCN under protic conditions. The presence of the co-oxidant K2S2O8 is crucial to this selectivity, probably by promoting oxidative transmetalation and the resulting formation of the Cu(iii)(acetylide)(CN) intermediate. All the reagents and salts used are commercially available, cheap and nontoxic, avoiding the use of highly toxic cyanide salts typically required in cyanation studies. The scope of this reaction is demonstrated with a set of alkynes and alpha-cyanoesters. The application of this method to late-stage functionalization of the terminal alkyne group in an estrone derivative is also feasible, showing its practical value for drug design.

A comparative study on mechanical and rheological properties of ternary rubber blends

Polymer blends lead to producing a new class of plastics, which may have better properties than being individual. In this study, the ternary blends of natural rubber (NR)/styrene-butadiene rubber (SBR)/nitrile rubber (NBR) were prepared via melt compounding, and then the physicomechanical properties of ternary NR/SBR/NBR blends were investigated. NR content was kept constant at 30 phr, while SBR and NBR fractions were varied simultaneously. The effect of the addition of 3 phr of ultrablend 4000 as compatibilizer on the compatibility of the rubber blends was studied. The results revealed that the use of the compatibilizer has significantly resulted in the clear stability of the scorch time and the optimum cure time of the blends. Tensile strength; stress at 100%, 200%, and 300% elongations; elongation at break %; compression strength; and compression set increase with the NBR content. There was a dramatic rise in the value of swelling ratio with the decrease of NBR. Moreover, the dynamic mechanical analysis showed some shifts in glass transition temperatures for blends to higher and lower temperatures as a function of the composition, which indicates the partial miscibility between the contributing components. Scanning electron microscopy of the tear fracture surfaces indicated that incorporation of NBR in the blends resulted in better adhesion of ternary blends and improved the tensile properties of ternary blends.

Synthesis and evaluation of the anti-proliferative activity of diaryl-3-pyrrolin-2-ones and fused analogs

Analogs containing a central 3-pyrrolin-2-one core with different methoxyphenyl and/or indole substituents were prepared and tested for anti-proliferative activity in U-937 cells. The most efficacious analogs were non-rigid, (non-fused) contained methoxyaryl groups located at the 4-position, and contained either methoxyaryl or indole groups located at the 3-position. Both the number of methoxy groups contained in the substituents and the particular location of the indole rings with respect to the lactam carbonyl had significant affects on anti-proliferative activity. Suzuki-Miyaura Arylations of Tetramic Acid Sulfonates: Evaluation of Lactam Protection, Sulfonate Esters, and Sterics

The synthesis of 3,4-diaryl-3-pyrroline-2-ones and 4-aryl-3-pyrrolin-2-ones using Suzuki-Miyaura cross-coupling reactions of tetramic acid sulfonates with arylboronic acids has been studied. The effect that sulfonate ester, sterics, and lactam protection has on the cross-coupling reaction was evaluated. As expected, triflates were better cross-coupling partners than the corresponding tosylates. The yields were only partially affected by the incorporation of aryl groups at the 3-position. On the intrinsic reactivity of highly potent trypanocidal cruzain inhibitors

The cysteine protease cruzipain is considered to be a validated target for therapeutic intervention in the treatment of Chagas disease. Hence, peptidomimetic cruzipain inhibitors having a reactive group (known as warhead) are subject to continuous studies to discover novel antichagasic compounds. Here, we evaluated how different warheads for a set of structurally similar related compounds could inhibit the activity of cruzipain and, ultimately, their trypanocidal effect. We first investigated in silico the intrinsic reactivity of these compounds by applying the Fukui index to correlate it with the enzymatic affinity. Then, we evaluated their potency against T. cruzi (Y and Tulahuen strains), which revealed the reversible cruzain inhibitor Neq0656 as a better trypanocidal agent (ECY50Y.strain = 0.1 mu M; SI = 58.4) than the current drug benznidazole (EC50Y.strain = 5.1 mu M; SI > 19.6). We also measured the half-life time by HPLC analysis of three lead compounds in the presence of glutathione and cysteine to experimentally assess their intrinsic reactivity. Results clearly illustrated the reactivity trend for the warheads (azanitrile > aldehyde > nitrile), where the aldehyde displayed an intermediate intrinsic reactivity. Therefore, the aldehyde bearing peptidomimetic compounds should be subject for in-depth evaluation in the drug discovery process.

The ability of halogenated triarylboranes to accept a lone pair of electrons from donor substrates renders them excellent Lewis acids which can be exploited as a powerful tool in organic synthesis. Tris(pentafluorophenyl)borane has successfully demonstrated its ability to act as a metal-free catalyst for an ever-increasing range of organic transformations. Herein we report the N-alkylation reactions of a wide variety of amine substrates including diarylamines, N-methylphenyl amines, and carbazoles with aryl esters using catalytic amounts of B(C6F5)(3). This mild reaction protocol gives access to N-alkylated products (35 examples) in good to excellent yields (up to 95%). The construction of a C-N bond at the propargylic position has also been demonstrated to yield synthetically useful propargyl amines. On the other hand, unsubstituted 1H-indoles and 1H-pyrroles at the C3/C2 positions afforded exclusively C-C coupled products. Extensive DFT studies have been employed to understand the mechanism for this transformation.
