Clarification of the USP compendial procedure for phenoxybenzamine hydrochloride via updating impurity profiles

The current United States Pharmacopeia-National Formulary (USP-NF) and the British Pharmacopoeia phenoxybenzamine (PBA) hydrochloride drug substance and drug product monographs describe an HPLC procedure for the determination of a specified impurity tertiary amine phenoxybenzamine and use the resolution of an unknown related substance from PBA as a system suitability criterion; however, neither structural information of the unknown related substance is provided nor reference standards of the two impurities are available. The ambiguity in pharmacopeias poses difficulties in implementing the procedure for quality control. To clarify the degradation pathways, and incorporate the impurity profile of PBA into the USP monographs, the degradation of PBA was revisited. PBA undergoes rapid degradation in neutral or basic aqueous solutions to generate the tertiary amine phenoxybenzamine as the predominant degradation product, which was confirmed as phenoxybenzamine hydroxide (PBA-OH). In addition, the unknown related substance was proposed as the phenoxybenzamine nitrile (PBA-CN) on the basis of LC-MS studies. The identity of PBA-CN was unambiguously verified via chemical synthesis, HPLC and NMR analyses. A stability-indicating method was developed and validated for the determination of PBA and its impurities, and was used to support USP monograph modernization. (C) 2020 Elsevier B.V. Experimental study on the influence of locked-in stress on the uniaxial compressive strength and elastic modulus of rocks

The rock contains many inclusions which produce high locked-in stress under the ground stress. In order to study the influence of locked-in stress on the mechanical properties of rocks, the rock-like materials and nitrile rubber particles are used to make a test block of the rock-like model which contains inclusions. The rubber particles will expand as the test block is heated, which creates locked-in stress in the inclusions. Uniaxial compression tests of similar model blocks with different locked-in stresses and different inclusion contents were performed by using a water bath and MTS-5T uniaxial compression testing machine. The results show that the peak strength and elastic modulus decreased with the increasement of locked-in stress and inclusion content. In the meantime, the relationship among the peak strength, the elastic modulus of the test piece, the locked-in stress and the inclusion content were obtained with the help of a mathematical fitting analysis of the quantitative formula. Furthermore, the expression and value curve of the joint impact factor are calculated. This paper evaluates the importance of the locked-in stress in the mechanical properties of the rock-like material and provide a guide for other researchers to further investigate the locked-in stress in rocks.

Triboelectric nanogenerator (TENG) and its self-powered sensor based on the principles of contact electricity generation and electrostatic induction have important application prospects in the fields of new energy and internet of things (IoT). In the contact separation process of polymer materials with different electronegativity values, due to the transfer of electrons, a changing electrostatic field will be generated in the space around the polymer. In the existing TENG research, the field strength perpendicular to the plane of the friction layer and the electrode layer is mainly used to generate electrostatic induction, and the electric field effect around the polymer is ignored. According to the principle of electrostatic induction, the internal charge of the conductor in the electric field will be redistributed, which provides a way for the conductor to generate an induced electrical signal on the surface of the conductor without contacting the friction material. In this paper, we design a non-contact triboelectric nanogenerator (NC-TENG) based on changing electrostatic field. The influence of the distance between the conductor and the friction material, the induction area of the conductor and the position of the conductor relative to the friction material on the induced electrical output performance are studied when silicone rubber and nitrile rubber are used as a friction material. The results show that the NC-TENG can produce a stable electrical signal output when the conductor is completely separated from the friction material. The induced voltage of NC-TENG decreases with the increase of the distance between the conductor and the friction material, and gradually increases with the increase of the conductor’s induction area. For the friction material with a size of 30 mm x 30 mm, the electrical output of NC-TENG tends to be stable when its conductor area is 60 mm x 45 mm. In addition, the different orientation of the conductor relative to the friction material also has a significant effect on the induced electrical output. The NC-TENG designed in this paper provides a novel electrical output generation mode, which provides a higher possibility for the subsequent research on TENG and the application of self-powered sensors.

Switchable Synthesis of Pyrroles and Pyrazines via Rh(II)-Catalyzed Reaction of 1,2,3-Triazoles with Isoxazoles: Experimental and DFT Evidence for the 1,4-Diazahexatriene Intermediate

4-Aminopyrrole-3-carboxylates and pyrazine-2-carboxylates were synthesized from 5-alkoxyisoxazoles and 1-sulfonyl-1,2,3-triazoles by tuning the Rh(II) catalyst and the reaction conditions. The reaction in chloroform at 100 degrees C under Rh-2(OAc)(4) catalysis provides 4-aminopyrrole-3-carboxylates in good yields. The use of Rh-2(Piv)(4) in refluxing toluene results in the formation of 1,2-dihydropyrazine-2-carboxylates as the main products, which can be converted by a one-pot procedure to pyrazine-2-carboxylates by heating with catalytic amounts of TsOH. According to the NMR and DFT investigations of the reaction mechanism, pyrroles and dihydropyrazines are formed, respectively, via 1,5- and 1,6-cyclization of common (5Z)-1,4-diazahexa-1,3,5-triene intermediates. The influence of the nature of the catalyst on the product distribution is rationalized in terms of the Rh-catalyzed isomerization of a pyrrolin-2-ylium-3-aminide zwitterion, the primary product of 1,4-diazahexatriene 1,5-cyclization.
