Solubility measurement, model evaluation and Hansen solubility parameter of piperonylonitrile in four binary solvents

The present research work proposed the mole fraction solubility and mixing thermodynamic properties of piperonylonitrile in different binary solvent mixtures such as ethanol + (acetone, 1,4-dioxane, DMF and methyl acetate) at T = 278.15 K to 323.15 K and p = 0.1 MPa. The results indicated that as the temperature and mass fraction of positive solvents increased, the measured solubility of piperonylonitrile increased in all mixtures considered. Analysis using Hansen solubility parameter revealed that the affinity between piperonylonitrile and all mixed solvents was related to multiple factors. The experimental solubility of piperonylonitrile in four binary solvent mixtures was regressed by employing four thermodynamic models such as UNIQUAC, Margules, Wilson and NRTL model. It was found that the Wilson model was accurate enough to correlate the experimental solubility. Moreover, the mixing thermodynamic properties of piperonylonitrile in different mixed solvents were also estimated and discussed according to the Wilson model and experimental data. The results of the negative Delta(mix)G values and positive Delta S-mix values demonstrated the entropy-driven and spontaneous mixing process of piperonylonitrile in all binary solvent mixtures. Low-temperature catalytic conversion of alkaline sewage sludge bio-oil to biodiesel: Product characteristics and reaction mechanisms

Sewage sludge is a promising biomass alternative for the production of liquid biofuels. In this study, bio-oil derived from the fast pyrolysis of domestic sewage sludge was upgraded in a micro Robinson-Mahoney reactor at 4 MPa and 260 degrees C, and in the presence of hybridized Ni/HZSM-5 catalyst and ethanol. Fourier transform infrared and gas chromatography-mass spectrometry analyses were used to identify the functional groups and chemical components of raw and upgraded sewage sludge bio-oil. Results showed that catalytic upgrading reduced the relative content of acids, aldehydes, phenols, ketones, and nitrogenated compounds in the pyrolytic bio-oil. Thus, the main components of the upgraded bio-oil product included esters (48.59%), alkanes (10.12%), aromatics (9.38%), and alkenes (5.08%). Water and nitriles in the raw pyrolytic bio-oil together with in-situ-produced carbon oxides helped in the esterification of raw bio-oil. Reaction mechanisms such as nitrile hydrolysis, hydrogenation, and esterification were proposed to elucidate the conversion of the non-acidic pyrolytic bio-oil into ester-dominating upgraded bio-oil. Overall, upgrading sewage sludge pyrolytic oil is crucial to produce high-quality diesel-range biofuel. (C) 2020 Elsevier B.V. All rights reserved.

Characterization of amino acid on glutathione adducts of cis-2-butene-1,4-dial, a reactive metabolite of furan

Metabolic activation of the hepatocarcinogen furan yields metabolites that react covalently with proteins, cis-2-Butene-1,4-dial is a microsomal metabolite of furan, This reactive aldehyde is thought to be the toxic metabolite that is responsible for the carcinogenic activity of furan. In order to characterize the chemistry by which this unsaturated dialdehyde could alkylate proteins, the products formed upon reaction of cis-2-butene-1,4-dial with model nucleophiles in PH 7.4 buffer were investigated, N-alpha-Acetyl-L-cysteine (AcCys) reacts with cis-2-butene-1,4-dial to form N-substituted pyrrolin-2-one adducts. N-Acetyl-L-cysteine (AcCys) reacts rapidly with cis-2-butene-1,4-dial to form multiple uncharacterized products. The inclusion of AcLys in this reaction mixture yielded an N-substituted 3-(S-acetylcysteinyl)pyrrole adduct which Links the two amino acid residues. Related compounds were isolated when cis-2-butene-1,4-dial and glutathione (GSH) were combined. In this case, cis-2-butene-1,4-dial cross-linked two molecules of GSH resulting in either cyclic or acyclic adducts depending on the relative GSH concentration. Incubation of furan with rat liver microsomes in the presence of [glycine-2-H-3]GSH led to the formation of radioactive peaks that coeluted with synthetic standards for the bisgluthathione conjugates. These studies demonstrate that the reactive cis-2-butene-1,4-dial formed during the microsomal oxidation of furan reacts rapidly and completely with amino acid residues to farm pyrrole and pyrrolin-2-one derivatives. Therefore, this metabolite is a likely candidate for the activated furan derivative that binds to proteins. The ease with which cis-2-butene-1,4-dial cross-links amino acids suggests that pyrrole-thiol cross-links may be involved in the toxicity observed following furan exposure.

Discovery of 3-hydroxy-3-pyrrolin-2-one-based mPGES-1 inhibitors using a multi-step virtual screening protocol

Targeting microsomal prostaglandin E-2 synthase-1 (mPGES-1) represents an efficient strategy for the development of novel drugs against inflammation and cancer with potentially reduced side effects. With this aim, a virtual screening was performed on a large library of commercially available molecules using the X-ray structure of mPGES-1 co-complexed with a potent inhibitor. Combining fast ligand-based shape alignment, molecular docking experiments, and qualitative analysis of the binding poses, a small set of molecules was selected for the subsequent steps of validation of the biological activity. Compounds 2 and 3, bearing the 3-hydroxy-3-pyrrolin-2-one nucleus, showed mPGES-1-inhibitory activity in the low micromolar range. These data highlighted the applicability of the reported virtual screening protocol for the selection of new mPGES-1 inhibitors as promising anti-inflammatory/anti-cancer drugs.

Volatile Organic Compounds (VOCs) of Endophytic Fungi Growing on Extracts of the Host, Horseradish (Armoracia rusticana)

The interaction between plant defensive metabolites and different plant-associated fungal species is of high interest to many disciplines. Volatile organic compounds (VOCs) are natural products that are easily evaporated under ambient conditions. They play a very important role in inter-species communication of microbes and their hosts. In this study, the VOCs produced by 43 different fungal isolates of endophytic and soil fungi during growth on horseradish root (Armoracia rusticana) extract or malt extract agar were examined, by using headspace-gas chromatography-mass spectrometry (headspace-GC-MS) and a high relative surface agar film as a medium. The proposed technique enabled sensitive detection of several typical VOCs (acetone, methyl acetate, methyl formate, ethyl acetate, methyl butanol isomers, styrene, beta-phellandrene), along with glucosinolate decomposition products, including allyl cyanide and allyl isothiocyanate and other sulfur-containing compounds-carbon disulfide, dimethyl sulfide. The VOC patterns of fungi belonging to Setophoma, Paraphoma, Plectosphaerella, Pyrenochaeta, Volutella, Cadophora, Notophoma, and Curvularia genera were described for the first time. The VOC pattern was significantly different among the isolates. The pattern was indicative of putative myrosinase activity for many tested isolates. On the other hand, endophytes and soil fungi as groups could not be separated by VOC pattern or intensity.

Theoretical investigation of protonated thiophene and two of its nitrile substituted derivatives (2-cyanothiophene and 3-cyanothiophene)

Theoretical and experimental spectroscopic data for protonated cyano-thiophenes (R-CNH+ with R = C4H3S), which are needed for their interstellar search and/or detection, are still lacking in the literature. Considering the high abundance and reactivity of H-3(+) in the interstellar medium (ISM), a quantum chemical investigation on protonated thiophene and two of its nitrile-substituted derivatives (2-cyanothiophene and 3-cyanothiophene) is undertaken for their characterization. The geometrical structures for the title species are calculated at the M06-2X/6-31G(d,p) level of theory, followed by an empirical correction for systematic errors. At the same level of theory, IR and Raman spectra are explored and the rotational parameters are calculated. The proton affinity (PA) of R-CN and the enthalpy, entropy and Gibbs free energy changes (Delta H-r, Delta S-r and Delta(r)G) of the reactions producing R-CNH+ are computed at the G2(MP2) and G3B3 levels of theory and at different temperatures. The PA calculations show that the protonation favors the nitrogen atom, while Delta H-r, Delta S-r, and Delta(r)G reveal the spontaneous reactions producing R-CNH+ and their neutral forms. In addition, quadrupole hyperfine structures are predicted, while the region where the brightest lines fall at different temperatures is discussed. These results are expected to assist astrophysicists and astrochemists in the search for new species in the ISM.

Poly(styrene)-co-2-vinylpyridine copolymer as a novel solid-phase adsorbent for determination of manganese and zinc in foods and vegetables by FAAS

The aim of this study is to extract zinc and manganese from foods and vegetables using an amphiphilic copolymer adsorbent, poly(styrene)-co-2-vinylpyridine which was synthesized via reversible addition fragmentation chain transfer (RAFT) polymerization from styrene and 2-vinyl pyridine in the presence of a trithiocarbonate and 2,2 ‘-azo-bis isobutyro nitrile (AIBN) in toluene solution under argon at 80 degrees C. Fourier Transform Infrared (FTIR) and proton nuclear magnetic resonance (H-1 NMR) spectroscopy were used in the characterization of the obtained copolymer. Under the optimum conditions, several validation variables such as uncertainty measurement, selectivity, robustness, precisions, matrix effects and accuracy were investigated. Taking an adsorption time of 15 min, detection limits of 0.04 mu g L-1 and 0.2 mu g L-1 and 7.9 mu g L-1 and enrichment factors of 145 and 110 were obtained for Mn(II) and Zn(II), respectively. The method was successfully applied to the analysis of Mn(II) and Zn(II) in foods and vegetables.
