A series of 1-(2-aminopropyl)-5-aryl-4-acyl-3-hydroxy-3-pyrrolin-2-ones were synthesized by three-component reactions of 4-substituted 2,4-dioxobutanoic acid methyl esters with mixtures of an aromatic aldehyde and 1,2-diaminopropane. Products of their reactions with hydrazine, p-phenetidine, and acetic anhydride were obtained. The analgesic, anti-inflammatory, and antimicrobial activities and hemostatic effect of several compounds were studied.

Synthesis and evaluation of the anti-proliferative activity of diaryl-3-pyrrolin-2-ones and fused analogs

Analogs containing a central 3-pyrrolin-2-one core with different methoxyphenyl and/or indole substituents were prepared and tested for anti-proliferative activity in U-937 cells. The most efficacious analogs were non-rigid, (non-fused) contained methoxyaryl groups located at the 4-position, and contained either methoxyaryl or indole groups located at the 3-position. Both the number of methoxy groups contained in the substituents and the particular location of the indole rings with respect to the lactam carbonyl had significant affects on anti-proliferative activity. This work provides a framework to better understand structure-activity relationships for inducing anti-proliferative activity in diaryl heterocyclic scaffolds. (C) 2016 Elsevier Ltd. Greener and regioselective ring opening of epoxides with TMSCN using potassium salts of magnetic carbon nitride

Identification of the C4 ‘-Oxidized Abasic Site as the Most Abundant 2-Deoxyribose Lesion in Radiation-Damaged DNA Using a Novel HPLC-Based Approach

A novel analytical high-performance liquid chromatography (HPLC)-based method of quantification of the yields of C4′-oxidized abasic sites, 1, in oxidatively damaged DNA has been elaborated. This new approach is based on efficient conversion of 1 into N-substituted 5-methylene-Delta(3)-pyrrolin-2-ones, 2, upon treatment of damaged DNA with primary amines in neutral or slightly acidic solutions with subsequent quantification of 2 by HPLC. The absolute and relative radiation-chemical yields of 1 in irradiated DNA solutions were re-evaluated using this method. The yields were compared with those of other 2-deoxyribose degradation products including 5-methylene-2(5H)-furanone, malondialdehyde, and furfural resulting from the C1′, C4′ and C5′-oxidations, respectively. The yield of free base release (FBR) determined in the same systems was employed as an internal measure of the total oxidative damage to the 2-deoxyribose moiety. Application of this technique identifies 1 as the most abundant sugar lesion in double-stranded (ds) DNA irradiated under air in solution (36% FBR). In single-stranded (ss) DNA this product is second by abundance (33% FBR) after 2-deoxyribonolactones (C1′-oxidation; 43% FBR). The production of nucleoside-5′-aldehydes (C5′-oxidation; 14% and 5% FBR in dsDNA and ssDNA, respectively) is in the third place. Taken together with the parallel reaction channel that converts C4′-radicals into malondialdehyde and 3′-phosphoglycolates, our results identify the C4’-oxidation as a prevalent pathway of oxidative damage to the sugar-phosphate backbone (50% or more of all 2-deoxyribose damages) in indirectly damaged DNA. Characterization of Dielectric Relaxation Process by Impedance Spectroscopy for Polymers: Nitrile Butadiene Rubber and Ethylene Propylene Diene Monomer

We invented a dispersion analysis program that analyzes the relaxation processes from dielectric permittivity based on a combination of the Havriliak-Negami and conductivity contribution functions. By applying the created program to polymers such as nitrile butadiene rubber (NBR) and ethylene propylene diene monomer (EPDM), several relaxation processes were characterized: an alpha process due to segmental motions of the C-C bond, an alpha ‘ process attributed to fluctuations in the end-to-end dipole vector of the polymer chain, the conduction contribution by the filler observed above room temperature, and secondary relaxation processes beta and gamma of motion for the side group in NBR. In the EPDM specimen, the beta process associated with the rotational motion of the side groups, the alpha process associated with the relaxation of local segmental motion, and the alpha beta process associated with the origin of the beta process at high temperatures above 305 K were observed. The Maxwell-Wagner-Sillars effect and conduction contribution were also presented. The molecular chains responsible for the relaxation processes were assigned by building molecular models of the two polymers. The temperature dependence of the relaxation strength and the shape parameters that characterize the process were investigated. From the temperature-dependent relaxation analysis, the merged alpha beta process, activation energy, and glass transition temperature were determined and compared.

Tribological properties of hydrogenated nitrile rubber in confrontation with hard particles under different axial loads

The influence of different axial loads on the tribological properties of Hydrogenated Nitrile Rubber (HNBR) under oil-based mud containing hard particles is studied through experimental and theoretical approaches. Friction and wear tests were performed by using pin-on-disc friction pairs. The findings indicate that the tribological properties of HNBR vary with axial load and wear time. The total friction coefficient can be divided into an adhesion term, a hysteresis term and an abrasive term through the analysis of tribological mechanism. The hysteresis term is due to micro-strain on the HNBR surface and overall deformation of the HNBR specimen. The variation of tribological properties can be well explained through the above theory and the hysteresis term is the main influence factor.

Uranium concentration using reactive polymer thin films for spectroscopic analyses

This contribution describes the development of reactive polymer films for the concentration of uranium from circumneutral pH solutions for spectroscopic analyses. These films were prepared by grafting uranium-selective polymers from polyethersulfone (PES) films via UV-initiated polymerization, and by introducing uranium-selective functional groups to polyacrylonitrile (PAN) films by chemical reaction. Ellipsometry was used to study poly(phosphoric acid 2-hydroxyethyl methacrylate ester) film growth kinetics on PES films. X-ray photoelectron spectroscopy of modified PAN films revealed conversion of nitrile groups to amidoxime groups to be as high as 40% and showed that the extent and depth of reaction could be varied precisely. Static uptake experiments with solutions of depleted uranium spiked with U-233 were conducted to determine uranium binding capacities and kinetics of the modified polymer films at different pH values from 4 to 8. Sorption isotherm data were fitted to the Langmuir model, and the highest sorption capacities of 1.09 x 10(-2) +/- 1.03 x 10(-3) mmol m(-2) and 1.02 x 10(-2) +/- 3.00 x 10(-3) mmol m(-2) were obtained at pH 6 for modified PAN (M-PAN) and PES (M-PES) films. Capacities at pH 4 and 8 were lower and could be explained by differences in sorption mechanisms. Uranium batch uptake kinetics followed a pseudo-second order rate model. Equilibrium uptake was attained within 3 h for M-PAN film and 1 h for M-PES film. Alpha spectroscopy pulse height spectra were analyzed to study the role of selective layer film thickness on peak energy resolution. Full width at half maximum values from 29 to 41 keV were recorded for M-PAN film and from 26 to 45 keV for M-PES film. Whereas uranium uptake increased with selective layer film thickness and varied with polymer chemistry/extent of modification, the peak energy resolution was independent of layer thickness and polymer chemistry within the experimental measurement uncertainties. Results from this work are being used to guide the development of thin-film composite membrane-based detection methods for the rapid, fieldable analysis of radionuclides in water for nuclear forensics investigations and environmental studies.

The SERS spectra were interpreted in terms of the structural changes of the tempyo labeled proteins adsorbed on the silver colloidal surface. The tempyo, spin label was found to be inactive in the Raman-RR and SERS spectra of the proteins. The a-helix conformation was concluded to be more favorable as the SERS binding site of bovine serum albumin. In the cytochrome c the enhancement of the bands assigned to the porphyrin macrocycle stretching mode allowed the supposition of the N-adsorption onto the colloidal surface.
