Ligand-controlled self-assembly of Ag(I) complexes with cyano-containing ligands and their tunable antimicrobial activities

Three cyano-containing ligands with varied size and shape, i.e. 1,4-dicyano benzene (1,4-dcb), 4,4′-biphenyldicarbonitrile (bpcn) and p-phenylenediacetonitrile (p-phda), were selected as ligands to self-assemble with AgCF3COO and 4,4′-dihydroxyoctafluorobipheny (ofbp) affording five new compounds. Complexes 1-3 ([Ag-2(CF3COO)2(1,4-dcb)(S)]center dot 2ofbp center dot H2O, 1, S=benzene, 2, S=toluene 3, S= m-xylene) were synthesized by the same linear spacer (1,4-dcb) in different solvents, and they exhibit similar 2D networks assembled by 1,4-dcb ligand bridging with 1D chains composing of anions and different coordinating solvents. Complex 4 ([Ag-2(CF3COO)(2)(bpcn)(2)]center dot C6H6 center dot 2ofbp center dot H2O) was isolated by replacing 1,4-dcb to the longer linear spacer (bpcn) however it shows a quite different 1D chain structure from those of 1-3. Further changing ligand to p-phda resulted only compound c as a co-crystal of p-phda and of bp. These totally different crystal structures suggest the ligand-controlled self-assembly processes. The followed antimicrobial assay results demonstrate that all complexes have effective antimicrobial activities with broad spectra. They exhibit excellent antimicrobial activities with low MICs towards both Gram-negative bacteria, Gram-positive bacteria and yeast. Especially, they illustrate encouraging antibacterial activities against antibiotic resistant bacteria (P. aeruginosa). It’s found that these frameworks have discriminating capacities to release Ag+ ions in water sustainably. Complex 4 demonstrated the best antimicrobial efficacy among five complexes. It’s revealed that the ligand exchangeability plays an important role in the anti-microbial activities. The lower concentration of Ag+ ions release of complex 3 is attributed to its compact packing structure on the basis of structure-property discussions.

Nitronyl nitroxides (NitR) are a family of persistent radicals widely used in molecular magnetism and recently suggested as potential candidates for spintronic applications. In this paper we characterize by X-and W-band Electron Paramagnetic Resonance (EPR) spectroscopy the new radical S-4-(nitronyl nitroxide) benzyl ethanethioate (NitSAc) designed for assembling on Au surfaces. We determined the radical magnetic tensors and studied by X-band pulse EPR its spin relaxation behaviour in fluid and glassy solutions of toluene. A comparison with the well known nitroxide 3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-oxyl (CTPO) is afforded. The advantages of using NitSAc in technological applications are discussed on the basis of the slow spin relaxation demonstrated by this study.

Synthesis of 4-substituted azopyridine-functionalized Ni(II)-porphyrins as molecular spin switches

We present the synthesis and the spin switching efficiencies of Ni(II)-porphyrins substituted with azopyridines as covalently attached photoswitchable ligands. The molecules are designed in such a way that the azopyridines coordinate to the Ni ion if the azo unit is in cis configuration. For steric reasons no intramolecular coordination is possible if the azopyridine unit adopts the trans configuration. Photoisomerization of the azo unit between cis and trans is achieved upon irradiation with 505 nm (trans -> cis) and 435 nm (cis -> trans). Concurrently with the isomerization and coordination/decoordination, the spin state of the Ni ion switches between singlet (low-spin) and triplet (high-spin). Flow Cytometric Analysis of the Influence of Myosmine on the Cell Cycle

Myosmine (3-(1-pyrrolin-2-yl)pyridine) is an alkaloid found in tobacco as well as various staple foods, fruits and vegetables. Myosmine has recently been suspected to be a tobacco-independent carcinogenic source. Using cell flow cytometry, we have examined the influence of myosmine on the cell cycle of murine erythroleukemia (MEL) cells in vitro and have compared this with its effects on murine bone marrow cells in vivo. Myosmine at low concentrations inhibited cell proliferation dose dependently; while at concentrations close to 300 mu mol dm(-3) it acted in a cytostatic fashion, that is, it increased the percentage of cells in the S and G2/M phases. At doses of 350-400 mu mol dm(-3) myosmine induced apoptosis and the hypoploid fraction. In vivo intraperitoneal injection of mice with 100 mg/kg of myosmine resulted in a statistically significant increase in the percentage of cells in S phase; i.e. from 13.75 to 18.22%. The percentage of bone marrow cells in the G2/M phase increased from 6.79 to 8.93 % in treated mice compared to controls. Machine learning-based prediction of enzyme substrate scope: Application to bacterial nitrilases

Predicting the range of substrates accepted by an enzyme from its amino acid sequence is challenging. Although sequence- and structure-based annotation approaches are often accurate for predicting broad categories of substrate specificity, they generally cannot predict which specific molecules will be accepted as substrates for a given enzyme, particularly within a class of closely related molecules. Combining targeted experimental activity data with structural modeling, ligand docking, and physicochemical properties of proteins and ligands with various machine learning models provides complementary information that can lead to accurate predictions of substrate scope for related enzymes. Here we describe such an approach that can predict the substrate scope of bacterial nitrilases, which catalyze the hydrolysis of nitrile compounds to the corresponding carboxylic acids and ammonia. Each of the four machine learning models (logistic regression, random forest, gradient-boosted decision trees, and support vector machines) performed similarly (average ROC = 0.9, average accuracy = similar to 82%) for predicting substrate scope for this dataset, although random forest offers some advantages. This approach is intended to be highly modular with respect to physicochemical property calculations and software used for structural modeling and docking.

Overhauser dynamic nuclear polarization and molecular dynamics simulations using pyrroline and piperidine ring nitroxide radicals

The efficiency of Overhauser dynamic nuclear polarization (DNP) depends on the local dynamics modulating the dipolar coupling between the two interacting spins. By attaching nitroxide based spin labels to molecules and by measuring the H-1 DNP response of solvent water, information about the local hydration dynamics near the spin label can be obtained. However, there are two commonly used types of nitroxide ring structures; a pyrroline based and a piperidine based molecule. It is important to know when comparing different experiments, whether changes in DNP enhancements are due to changes in local hydration dynamics or because of the different spin label structures. In this study we investigate the key parameters affecting DNP signal enhancements for 3-carbamoyl-2,2,5,5-tetramethyl-3-pyrrolin-1-oxyl, a 5-membered ring nitroxide radical, and for 4-oxo-2,2,6,6-tetramethyl-1-piperidinyloxy, a 6-membered ring nitroxide radical. Using X-Band DNP, field cycling relaxometry, and molecular dynamics simulations, we conclude that the key parameters affecting the DNP amplitude of the H-1 signal of water to be equal when using either nitroxide. Synthesis and characterization of a crosslinked membrane based on sulfonated poly(aryl ether sulfone) and sulfonated polyvinyl alcohol applied in direct methanol fuel cells

Vinyl-terminated sulfonated poly (arylene ether sulfone) (VSPAES) and sulfonated polyvinyl alcohol containing vinyl group (VSPVA) have been prepared. The VSPVA and VSPAES were crosslinked by adding AIBN as initiator. The results of FT-IR and H-1-NMR indicated the existence of sulfonic acid groups and crosslinked structure in these crosslinked membranes. The microphase structures of the membranes are analyzed by SEM. The proton conductivities of membranes were above 10 mS cm(-1), which indicated that it basically meet the needs of fuel cells. The methanol permeability coefficients of CSPAES membrane is 2.8×10(-7) cm(2) s(-1), remarkably lower than neat SPAES (8.5×10(-7) cm(2) s(-1)) and Nafion117 (14.1×10(-7) cm(2) s(-1)). Moreover, the proton selecticity of CSPAES membrane is close to 2 times higher than that of Nafion 117 membrane. The crosslinked membrane showed better stability than the linear SPAES membrane.
